Staining technique: Periodic acid / schiff
Synonym(s): PAS
Introduction
- Stain reacts with structures containing a carbohydrate component (1:2 glucol grouping, including polysaccharides, glycoproteins and glycolipids).
- Periodic acid is the oxidant, others can be used then results may vary slightly.
Uses
- Identification of fungal hypae, fungal spores and yeasts.
- Demonstrates glycogen in cells, mucus in various cells and tissues, the basement membrane that underlies epithelia and reticular fibers in connective tissue.
Alternative techniques
- Gomori's methamine silver (stain wall of any living or dead fungi).
